Sorry if this sounds so obvious, but you never know. The camera must be switched on when you connect it to the PC. Sorry if you already knew this, but it tripped me up when I first got a digicam.

A couple of questions:
1. Has the camera worked with your PC in the past? If so, have you made any changes to the camera or PC since it was last functional?
2. Did you check the "Troubleshooting" guide in the back of the camera manual?
From the troubleshooting guide:
A. Make sure the USB item in the Setup menu is set to "Mass Storage" rather than PTP
B. Make sure the camera is registered in the Windows Device Manager
Another thought - maybe the cable has developed a fault. Can you try a different cable?
Good luck,
Richard Baker

Also if I may.... what Windows are you using .. if it's not XP... have to use "safely remove/connect hardware function" and as mentioned before, power off before connecting .. then on
Later

If you haven't properly disconnected you may have damaged the drivers, in which case you need to reload them either from your supplied software or many of them are available as free downloads from internet sites.
